Precision intradermal cancer vaccination with liposome and injection device integration (PRECIVAC)

This project brings together Leiden University Medical Center and Idevax to develop a new strategy for precision cancer vaccination. By combining advanced liposome-based vaccine formulations with a dedicated intradermal injection device, the partnership aims to improve how therapeutic cancer vaccines are delivered into the skin. The project focuses on generating robust preclinical data in ex vivo human skin models and laboratory immune assays.

Cancer remains one of the leading causes of death worldwide. While immunotherapy has transformed treatment for several cancers, many approaches rely on systemic administration, which can lead to side effects and variable effectiveness. The skin is rich in specialised immune cells that can initiate strong anti-tumour responses when properly activated. Delivering vaccines directly into the skin offers the opportunity to stimulate immunity more precisely, potentially improving efficacy while limiting unwanted systemic effects. Innovation in both formulation and delivery technology is therefore essential.

The project investigates how peptide-containing liposomes can be optimally delivered using a clinically validated intradermal injection device. First, compatibility between vaccine formulations and the device will be assessed. Next, delivery performance will be evaluated in ex vivo human skin to study biodistribution and fluid migration. Finally, immune activation will be analysed using established laboratory assays. This integrated approach allows systematic optimisation of both formulation and delivery parameters.

The main deliverables include validated compatibility data between liposomal vaccines and the injection device, detailed insight into skin distribution patterns, and preclinical evidence of local immune activation. These results will provide a solid foundation for further development toward future clinical application.
