
Hibernation-based treatment for Alzheimer’s disease
SUL-138 enhances mitochondrial function to mitigate cognitive decline and amyloid burden in Alzheimer's disease
This collaborative project, involving Amsterdam UMC, VU, UMCG, and Sulfateq B.V., investigates SUL-138, a compound derived from hibernation, for its potential to restore mitochondrial function and alleviate Alzheimer's disease symptoms. The preclinical studies of this consortium among others demonstrate that SUL-138 improves memory, reduces amyloid plaque load, and enhances synaptic activity by shifting mitochondrial metabolism towards fatty acid oxidation, offering a promising therapeutic avenue. These findings have contributed to the advancement of SUL-138 into Phase 1 clinical trials in healthy volunteers and preparations for a Phase 2 clinical trial in AD patients, marking a significant step towards clinical application.
Alzheimer's disease is a significant global health challenge. The number of patients is projected to rise to 14 million by 2030, leading to an estimated €250 billion in treatment and care costs. Currently, no cure exists, and available treatments offer limited benefits. This underscores the urgent need for innovative therapies targeting the underlying disease mechanisms.
The project focuses on SUL-138's effects on mitochondrial function, aiming to restore energy production in neurons and prevent neurodegeneration. By utilizing both animal models and human neuronal cells, the research provides a comprehensive understanding of the compound's potential therapeutic impact and mechanisms of action.
Preliminary results indicate that SUL-138 enhances mitochondrial activity, improves neuronal health, and mitigates Alzheimer's-related symptoms. These findings support the continued development of SUL-138 as a promising candidate for Alzheimer's treatment and have contributed to its progression into Phase 1 clinical trials. The partners of this successful consortium remain fully committed to advancing new treatments for neurodegenerative disorders.
